  • Fold Description: The only structural template for Pfam family PF02829 and COG family COG1827 (3H domain). Transcriptional regulator from biotin repressor family, no paralogs, several (about 20) homologs in bacteria. Covered by two PFAM domains, the N-terminal 60 residues by an iron-dependent repressor DNA binding domain, the C-terminal domain by 3H domain, a small molecule binding domain (hypothetical function). Structure of the first domain is easily predicted as a classical HTH DNA binding motif and with strong similarity to multiple PDB entries (~2 angstrom rmsd), in line with moderate sequence similarity (~30% id). The second domain has a completely unexpected structural similarity to phosphocarrier protein (PDB:1qr5) (3.5 angstrom rmsd over 80 amino acids) that was not predicted by any public fold prediction programs (8% sequence identity). The functional implications of this similarity is not clear, as none of the functionally important residues in the phosphocarrier protein are conserved.
